Yes, you can. But you should apply it to a small, inconspicuous area of your flooring for testing purposes before proceeding with the full application. Make sure that the surface of the floor is clean of all dirt and dust. If you are using a buffer, then mop the pad with the floor cleaner using the same method as you would use to mop your floor with water. It is necessary to wipe off the excess cleaner before buffing. You can use Mop & Glo on vinyl plank flooring. But, be sure to protect your hands when you use it. You will need to wear rubber gloves and an apron or dust mask. Make sure there is no one else in the room when you use the product.
